ABSTRACT

Lung is susceptible to external disturbance, resulting in a variety of acute and chronic lung diseases. Functionalized nanoparticles as carriers can carry drugs through multiple biological barriers of lung into lung lesions, but there are some problems such as poor targeting and low therapeutic efficiency. As a drug carrier, membrane-coated biomimetic nanoparticles have the characteristics of immune system escape, active targeting, inflammatory chemotaxis and crossing physiological barriers due to the retention of the characteristics of the source cells. Therefore, it has been widely used in the treatment of lung diseases in recent years. In this review, the application of membrane-coated biomimetic nanoparticles in the treatment of lung diseases in the recent years was summarized and classified. Cell membrane sources include erythrocyte membrane, platelet membrane, macrophage membrane, neutrophil membrane, lung epithelial membrane, lung surfactant, endothelial membrane, cancer cell membrane, bacterial membrane, hybrid membrane and so on. The purpose of this review is to provide a new idea for treating lung diseases with membrane-coated biomimetic nanoparticles.

ABSTRACT

As confusion mounts over RNA isoforms involved in phenotypic plasticity, aberrant CpG methylation-mediated disruption of alternative splicing is increasingly recognized as a driver of intratumor heterogeneity (ITH). Protease serine 3 (PRSS3), possessing four splice variants (PRSS3-SVs; PRSS3-V1-V4), is an indispensable trypsin that shows paradoxical effects on cancer development. Here, we found that PRSS3 transcripts and their isoforms were divergently expressed in lung cancer, exhibiting opposing functions and clinical outcomes, namely, oncogenic PRSS3-V1 and PRSS3-V2 versus tumor-suppressive PRSS3-V3, by targeting different downstream genes. We identified an intragenic CpG island (iCpGI) in PRSS3. Hypermethylation of iCpGI was mediated by UHRF1/DNMT1 complex interference with the binding of myeloid zinc finger 1 (MZF1) to regulate PRSS3 transcription. The garlic-derived compound diallyl trisulfide cooperated with 5-aza-2'-deoxycytidine to exert antitumor effects in lung adenocarcinoma cells through site-specific iCpGI demethylation specifically allowing MZF1 to upregulate PRSS3-V3 expression. Epigenetic silencing of PRSS3-V3 via iCpGI methylation (iCpGIm) in BALF and tumor tissues was associated with early clinical progression in patients with lung cancer but not in those with squamous cell carcinoma or inflammatory disease. Thus, UHRF1/DNMT1-MZF1 axis-modulated site-specific iCpGIm regulates divergent expression of PRSS3-SVs, conferring nongenetic functional ITH, with implications for early detection of lung cancer and targeted therapies.

ABSTRACT

The pharmaceutical manufacturing model is gradually changing from intermittent manufacturing to continuous manufacturing and intelligent manufacturing. This paper briefly reviewed the supervision and research progress in continuous pharmaceutical manufacturing in China and abroad and described the definition and advantages of continuous pharmaceutical manufacturing. The continuous manufacturing of traditional Chinese medicine(TCM) at the current stage was summarized in the following three terms: the enhancement of the continuity of intermittent manufacturing operations, the integration of continuous equipment to improve physical continuity between units, and the application of advanced process control strategies to improve process continuity. To achieve continuous manufacturing of TCM, the corresponding key technologies, such as material property characterization, process modeling and simulation, process analysis technology, and system integration, were analyzed from the process and equipment, respectively. It was proposed that the continuous manufacturing equipment system should have the characteristics of high speed, high response, and high reliability, "three high(H~3)" for short. Considering the characteristics and current situation of TCM manufacturing, based on the two dimensions of product quality control and production efficiency, a maturity assessment model for continuous manufacturing of TCM, consisting of operation continuity, equipment continuity, process continuity, and quality control continuity, was proposed to provide references for the application of continuous manufacturing technology for TCM. The implementation of continuous manufacturing or the application of key continuous manufacturing technologies in TCM can help to systematically integrate advanced pharmaceutical technology elements and promote the uniformity of TCM quality and the improvement of production efficiency.

@#[摘 要] 目的:系统评价基于PD-1/PD-L1抑制剂的免疫联合治疗(以下称“免疫联合治疗”)对比舒尼替尼治疗晚期肾细胞癌(RCC)的安全性和有效性。方法:检索PubMed、Embase、Cochrane Library及中国知网(CNKI)数据库,收集国内外公开发表的免疫联合治疗对比舒尼替尼应用于晚期RCC的随机对照试验(RCT),检索时间均为自建库时间至2022年10月。由两名研究者独立评价纳入研究的质量、提取资料并交叉核对,采用StataMP16.0软件进行Meta分析。结果:共纳入6项RCT,Meta分析结果显示,(1)有效性:与舒尼替尼相比,免疫联合治疗显著提高了晚期RCC患者的总生存期[OS,HR=0.74,95% CI (0.67,0.80),P<0.01]和无进展生存期[PFS,HR=0.66,95% CI (0.51,0.81),P<0.01];(2)安全性:两治疗组均有较高的不良反应(AE)发生率,差异无统计学意义。但免疫联合治疗组发生皮肤及内分泌系统AE显著高于舒尼替尼治疗组,而血液系统相关AE则明显低于舒尼替尼治疗组;(3)以1%为临界点,免疫联合治疗组的RCC患者,无论是PD-L1阳性或阴性的,其OS和PFS均高于舒尼替尼组。结论:免疫联合治疗可显著延长晚期RCC患者的OS和PFS,但不同系统发生AE有差异,且RCC患者PD-L1表达状态(1%为临界点)并不影响免疫联合治疗的获益。

ABSTRACT

ObjectiveTo investigate the compatibility rule of traditional Chinese patent medicines (TCPMs) against liver diseases through network analysis. MethodWith “liver” as the search term, TCPMs against liver diseases were retrieved from volume Ⅰ of Chinese Pharmacopoeia (2020 edition), and the basic information of them was collected. TCPMs with same Chinese medicinal materials (CMMs), usage, and indications, but different dosage forms, were unified as one formula. Mutual information entropy (MIE) of CMM couples was calculated to quantify the relationship between them, and the top 25% CMM pairs in MIE were used to construct the compatibility network, with CMM as node and the relationship between CMM pairs as the edge. Key CMM and frequently used CMM combinations were identified based on node centrality and cluster analysis, respectively. The indications of TCPMs related to the CMMs in clusters were recorded. Cytoscape 3.6.1 was employed for visualization and topology analysis of the compatibility network. ResultA total of 179 TCPMs, involving 428 CMMs, were retrieved. Angelicae Sinensis Radix, Paeoniae Radix Alba, and Glycyrrhizae Radix et Rhizoma were identified as key CMMs with high frequency, and Cuscutae Semen-Lycii Fructus, Citri Reticulatae Pericarpium-Cyperi Rhizoma, and Ecliptae Herba-Ligustri Lucidi Fructus combinations had high MIE. Furthermore, the CMMs were clustered into ten groups corresponding to different diseases which, however, all belonged to digestive diseases. ConclusionThis study unveils potential CMM pairs and common CMM combinations against liver diseases, which can serve as a reference for revealing compatibility rules of CMMs and research and development of Chinese medicine.

ABSTRACT

Malocclusion is one of the three most common oral diseases reported by World Health Organization(WHO). In China, its incidence rate is rising. Malocclusion seriously affects the dental and maxillofacial function, facial appearance and growth development of nearly 260 million children in China, and what is more, it affects their physical and mental health development. Malocclusion occurrence is related to genetic and environmental factors. Early treatment of malocclusion can create a good dental and maxillofacial development environment, correct abnormal growth and control the adverse effects of abnormal genetic factors. It can effectively reduce the prevalence of children's malocclusion and enhance their physical and mental health. This is an urgent need from the economic perspective of our society, so it has great practical and social significance. Experts from the project group "standard diagnose and treatment protocols for early orthodontic intervention of malocclusions of children" which initiated by China National Health Institute of Hospital Administration wrote the "China Experts' Consensus on Preventive and Interceptive Orthodontic Treatments of Malocclusions of Children", which aims to guide and popularize the clinical practice, improve the clinical theory and practice level, and accelerate the disciplinary development of early treatment of children's malocclusion in China. The consensus elaborates the harmfulness of malocclusion and the necessity of early treatment, and brings up the principles and fundamental contents. Based on the law of dental and maxillofacial development, this paper puts forward the guiding suggestions of preventive and interceptive treatments in different stages of dental development ranging from fetus to early permanent dentition. It is a systematic project to promote and standardize the early treatment of malocclusion. Through scientific and comprehensive stratified clinical practice and professional training, the clinical system of early treatment of malocclusion in China will eventually be perfected, so as to comprehensively care for children's dental and maxillofacial health, and improve their oral and physical health in China.

ABSTRACT

Textual research on Chinese herbs is the preliminary work for the preparation of famous classical prescriptions. Through literature review,it was found that the researches of Persicae Semen focused on chemical compositions,pharmacological mechanism and medical record analysis in the recent years,and few researches based on the ancient literature were about the origin,concocting methods,flavor,meridian tropism,effects and indications. Textual research shows that the most commonly used names are Taoren and Taoheren,with a wide range of producing areas. The plant origin of Persicae Semen has not changed much since ancient times. Before the Qing dynasty,the plant origin of Persicae Semen was from the seeds of a kind of fruit named Shantao or Maotao,and in modern times,the seeds of Amygdalus persica or A. davidiana have become the major source. While different books have different views on Latin names for the origin of the Persicae Semen. We suggest that the Latin names of A. persica and A. davidiana should be more reasonable for Tao and Shantao respectively .In the concocting methods of Persicae Semen for activating vital energy and blood circulation,raw Persicae Semen should be used with peel and tip,while for moisturizing dryness,it should be fried into yellow without peel. Therefore,in the concocting methods of Persicae Semen for Taohe Chengqitang and Taohong Siwutang,the raw materials should be fried into yellow without peel or tip,while for Shentong Zhuyutang,raw Persicae Semen materials should be used with peel and tip. The indications of Persicae Semen include amenorrhea,lump,parasite,obstruction of chest,cough and asthma,constipation,etc.,and the people with blood deficiency,blood dryness and the pregnant women should use it with caution or should not use it. The modern clinical application of Persicae Semen is only a partial inheritance of ancient literature,which means that the Persicae Semen still has many effects to be verified and studied,and it is worthwhile for further exploration in order to expand its clinical application. The records of ancient literature on flavor,meridian tropism,and quality evaluation of Persicae Semen were consistent with those in Chinese Pharmacopoeia. Because of the similar appearance,it is especially difficult to distinguish Persicae Semen and Armeniacae Semenis Amarum after crushing ,requiring much time and money for identification. It is recommended that medical institutions should purchase Persicae Semen without crushing as far as possible,then decide the best concocting methods according to the clinical requirement.

ABSTRACT

The Coronavirus disease 2019 (COVID-19) is an emerging infectious disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Within a matter of months, this highly contagious novel virus has led to a global outbreak and is still spreading rapidly across continents. In patients with COVID-19, underlying chronic diseases and comorbidities are associated with dismal treatment outcomes. Owing to their immunosuppressive status, patients with hematological malignancies (HMs) are at an increased risk of infection and have a worse prognosis than patients without HMs. Accordingly, intensive attention should be paid to this cohort. In this review, we summarize and analyze specific clinical manifestations for patients with coexisting COVID-19 and HMs. Furthermore, we briefly describe customized management strategies and interventions for this susceptible cohort. This review is intended to guide clinical practice.

The research and development of classical famous prescriptions is an important way to actively promote the development of traditional Chinese medicine.It is particularly important to sort out the historical evolution of the composition of traditional famous prescriptions to make clear the origin,producing areas and concocting methods of traditional Chinese medicine,which is the source of ensuring the safety and efficacy.Through literature review,it is found that at present,the research of mulberry white skin focuses on chemical composition,pharmacological mechanism and modern clinical research,and there are few ancient literature studies.Therefore,based on the ancient literature,the author conducts a comprehensive textual research on mulberry bark from its name,origin,producing areas and concocting methods and other aspects,in order to provide literature reference for the research and development of the prescription involving mulberry bark in the classic prescription.Through research,we can know that there are more than 20 aliases of mulberry bark,and the most commonly used names in modern times are "sangbaipi""sanggenbaipi""sangpi",etc.In Tang and Song dynasties and before,mulberry bark was mainly composed of Morus alba var. alba and jisang,after Tang and Song dynasties,mulberry bark plant sources showed diversity,in modern times,Morus alba var. alba was gradually identified as the main medicinal species of mulberry bark.Therefore,it is suggested that Morus alba var. alba be selected as the plant source of mulberry bark.According to ancient books,mulberry trees are cultivated everywhere,with Jiangsu,Zhejiang and Sichuan as the best areas,and Henan and Anhui as the most popular areas in modern times.The conclusion of ancient and modern quality of mulberry bark is basically the same,the root skin is white,thick and sweet.The morden concocting methods of mulberry bark mainly include raw mulberry bark,honey mulberry bark and fried mulberry bark.According to the textual research of ancient literature,in addition to the above three kinds of medical specifications,there have been concocting methods without auxiliary materials,such as burning,baking and roasting,as well as concocting methods with auxiliary materials,such as bran roasting,rice swill soaking,honey wine roasting,etc.The concocting methods of mulberry bark used by the classical famous recipe should be selected in combination with specific drug provisions.

@#BACKGROUND: The study aimed to evaluate the predictive role of interleukin-6 (IL-6) and chronic obstructive pulmonary disease (COPD) assessment test (CAT) score in mechanical ventilation (MV) in COPD patients at the acute exacerbation stage in the emergency department (ED). METHODS: For a one-year period, among adult patients in the ED who met the criteria of acute exacerbation of COPD, 158 who received MV within 48 hours after admission were compared to 294 who didn't require MV within the same period after admission. IL-6 level and CAT score were compared between the two groups. The predicted value of IL-6 and CAT score was assessed by logistic regression analysis and a receiver operating characteristic (ROC) curve. RESULTS: The IL-6 and CAT scores in the 158 MV patients were much higher than those without. IL-6 and CAT scores were independent predictors of MV within 48 hours using logistic regression analysis (IL-6: odds ratio [OR] 1.053, 95% confidence interval [CI] 1.039–1.067, P<0.001; CAT score: OR 1.122, 95% CI 1.086–1.159, P<0.001). The combination of IL-6 and CAT scores (area under ROC curve [AUC] 0.826, 95% CI 0.786–0.866, P<0.001) improved the accuracy of predicting MV within 48 hours when compared with IL-6 (AUC 0.752, 95% CI 0.703–0.800, P<0.001) and CAT scores alone (AUC 0.739, 95% CI 0.692–0.786, P<0.001). The sensitivity and specificity were 69.6%, 74.1%, 75.32% and 63.6%, respectively. CONCLUSION: The combined of IL-6 and CAT scores is useful for evaluating the risk of COPD patients at acute exacerbation in ED, and can provide a predictive value for MV or not within 48 hours.

ABSTRACT

Objective:To investigate the application value of renal depth correction by the integrated CT in glomerular filtration rate (GFR) determination by 99Tc m-diethylene triamine pentoacetic acid (DTPA) renal dynamic imaging for patients with hydronephrosis. Methods:A total of 338 patients (191 males, 147 females, age (49.6±14.5) years) in Beijing Tsinghua Changgung Hospital from April 2016 to June 2019 with different degrees of hydronephrosis were respectively analyzed. Patients were divided into groups of normal-mild, normal-moderate, normal-heavy, mild-mild, mild-moderate, mild-heavy, moderate-moderate, moderate-heavy and heavy-heavy according to the degree of bilateral hydronephrosis. The renal depth was measured by the integrated CT method and the routine method, and the absolute value of bilateral renal depth difference in normal-mild, normal-moderate and normal-heavy groups was calculated by the 2 methods. Based on the renal depth measured by the 2 methods, the single renal GFR was measured by 99Tc m-DTPA dynamic renal imaging Gates method and compared between the 2 methods. Total GFR measured by the 2 methods were compared with estimated GFR (eGFR). One-way analysis of variance analysis, paired t test, and Pearson correlation analysis were used. Results:For the integrated CT measurements, the absolute value of bilateral renal depth difference in normal-mild, normal-moderate and normal-heavy groups were significantly different ((0.39±0.24), (1.16±0.65) and (1.00±0.90) cm; F=15.241, P<0.05). The renal depth and the single renal GFR measured by the integrated CT method were higher than those measured by the routine method ( t values: 16.06-19.78, 14.27-17.23, all P<0.05) in the kidneys with normal, mild, moderate and heavy hydronephrosis. There were significant differences between the total GFR measured by the routine method and eGFR in all groups ( t values: from -8.178 to 5.879, all P<0.05); however, in the integrated CT method, except that the total GFRs in moderate-heavy group and heavy-heavy group were overestimated ( t values: 3.035 and 11.247, both P<0.05), there were no significant differences between the total GFR ((111.57±17.37), (103.71±15.22), (79.79±12.62), (100.33±18.49), (100.28±15.43), (84.09±20.72) and (74.14±14.57) ml·min -1·1.73 m -2) and eGFR ((109.16±12.81), (103.20±13.26), (78.60±14.12), (100.98±15.20), (99.89±14.05), (84.61±20.24) and (73.44±14.57) ml·min -1·1.73 m -2) in normal-mild, normal-moderate, normal-heavy, mild-mild, mild-moderate, mild-heavy and moderate-moderate groups ( t values: from -0.301 to 1.948, all P>0.05). The total GFR measured by the 2 methods were significantly correlated with eGFR in 338 patients with hydronephrosis ( r values: 0.888 and 0.928, both P<0.01). Conclusion:Compared with the routine method, except for the moderate-heavy group and heavy-heavy group, renal depth correction by the integrated CT may have greater clinical significance in GFR measurement by renal dynamic imaging for patients with hydronephrosis.

ABSTRACT

OBJECTIVE:To investigate the effects of gypenosides (GPs)on gene expression of major urinary proteins (Mups) in liver tissue of hypercholesterolemia model mice. METHODS :C57BL/6J mice were divided into control (ND)group,model (HFD)group and GPs therapy (GP)group according to body weight (BW),with 11 mice in each group. Except for ND group , other groups were given high-lipid diet to induce hypercholesterolemia model. From the 17th week of feeding ,ND group and HFD group were given constant volume of 0.1%CMC-Na solution intragastrically ;GP group were given GPs suspension (250 mg/kg) intragastrically,once a day ,for consecutive 22 weeks. BW ,the levels of blood glucose (BG)and blood lipid (TC,LDL-C)were detected in each group. Total RNA of liver tissue was extracted ,and reverse transcription library was constructed and RNA-seq sequencing was performed. The differentially expressed genes were screened by PCA ,volcano map and scatter plot. RT-qPCR was used for verification for differentially expressed genes. The correlation between the expression of differentially expressed genes and the above pharmacodynamic indexes was analyzed by bivariate analysis. RESULTS :Compared with ND group ,BW,the levels of BG,TC and LDL-C in HFD group were increased significantly (P<0.05). Compared with HFD group ,above indexes of GP group were decreased significantly except for BW (P<0.05). PCA showed that the data of ND group and HFD group distributed in different quadrants ,and the data distribution of GP group was between above two groups. mRNA of Mup4,Mup5,Mup11,Mup15 and Mup21 in liver tissue of mice were increased significantly after treated with high-fat diet (P<0.05). mRNA of Mup3,Mup4, Mup5,Mup8,Mup12 and Mup21 were decreased significantly after treated with GPs (P<0.05). In ND group vs. HFD group and HFD group vs. GP group ,mRNA of Mup4,Mup5 and Mup21 genes changed significantly and the trend was opposite. Results of RT-qPCR verification showed that compared with ND group ,relative mRNA expression of Mup4,Mup5 and Mup21 gene were increased significantly in HFD group (P<0.05). Correlation analysis revealed that mRNA expression of Mup5 was positively correlated with the levels of TC and BG (r=0.727 1,0.670 6,P<0.05),mRNA expression of Mup4 and Mup21 were positively correlated with the level of BG (r=0.737 8,0.721 5,P<0.05). CONCLUSIONS :GPs can regulate the expression of Mups genes in liver tissue of hypercholesterolemia model mice , and reduce glucose and lipid level through regulating the mRNA over-expression of Mup4,Mup5 and Mup21.

ABSTRACT

In order to better evaluate the transport effect of nanoparticles through the nasal mucosa, an nasal cavity-mimic model was designed based on M cells. The differentiation of M cells was induced by co-culture of Calu-3 and Raji cells in invert model. The ZO-1 protein staining and the transport of fluorescein sodium and dexamethasone showed that the inverted co-culture model formed a dense monolayer and possessed the transport ability. The differentiation of M cells was observed by up-regulated expression of Sialyl Lewis A antigen (SLAA) and integrin 1, and down-regulated activity of alkaline phosphatase. After targeting M cells with iRGD peptide (cRGDKGPDC), the transport of nanoparticles increased. , the co-administration of iRGD could result in the increase of nanoparticles transported to the brain through the nasal cavity after intranasal administration. In the evaluation of immune effect , the nasal administration of OVA-PLGA/iRGD led to more release of IgG, IFN-, IL-2 and secretory IgA (sIgA) compared with OVA@PLGA group. Collectively, the study constructed M cell model, and proved the enhanced effect of targeting towards M cell with iRGD on improving nasal immunity.

ABSTRACT

Objective@#To understand the behavioral characteristics of Chinese college students who have sex with men, and to provide a basis for more targeted health education.@*Methods@#A systematic search of studies related to the high-risk behaviors of MSM of Chinese college students published from January 1, 2015 to November 1, 2018 in the Chinese Journal Full-text database (CNKI), China Biology Medicine database(CBM), PubMed, VIP and Wanfang database was conducted, and relevant data was extracted for Meta-analysis.@*Results@#A total of 12 studies were included, there were 3 839 MSM of college students enrolled in theses study,the results showed that the overall or pooled prevalence of HIV in MSM of college students was 79.1%(95%CI=73.1%-85.0%) in recent years; the combined rate of the number of sexual partners≥2 was 58.9%(95%CI=46.5%-71.3%); the combined rate of condom use for gay sex in the last 6 months was45.3%(95%CI=37.9%-52.7%); the combined rate of condom use for per heterosexual sex was 43.6%(95%CI=29.6%-57.5%); the combined detection rate of HIV was 4.7%(95%CI=2.7%-6.8%).@*Conclusion@#The high-risk behavior of MSM among Chinese college students is prominent, and online dating is highly dependent. Therefore, colleges should strengthen the health education of prevention of AIDS.

ABSTRACT

Objective To explore the pathogenesis of bladder cancer from the molecular level and provide new ideas for clinical diagnosis and prognosis evaluation. Methods Human bladder cancer-associated gene chip data GSE31189, including 52 bladder cancer samples and 40 normal bladder samples, was downloaded from a database of gene chips, and then Morpheus (https://software.broadinstitute.org/morpheus/) online tools were used to analyze cancer urothelial tissue and normal urothelial tissue. Differentially expressed genes were then analyzed online using gene-cloud of biotechnology information(GOBI) (https://www.GCBI.com.cn) for enrichment of differentially expressed gene signaling pathways and differential gene interactions. Finally, differential selection genes were selected for cell counting kit-8(CCK-8) , invasion and Western blotting assays. Initially verify its function. Results According to the q value, the top 20 genes with the largest difference were selected. Among them, 18 genes were up-regulated and 2 genes were down-regulated in bladder cancer. Gene ontology(GO)analysis revealed that these differentially expressed genes are mainly involved in many biological functions such as inflammatory responses, immune responses, negative regulation of apoptosis, and transcriptional negative control from the RNA polymerase D promoter. Pathway analysis showed that these differentially expressed genes are involved in biological processes such as transcriptional dysregulation, metabolic pathways, and nuclear factor kappa beta (NF-kB) signaling pathways in cancer. Gene network analysis found that CXC chemokine receptor 4 (CXCR4) and mitogen-activated protein kinase 10 (MAPK10) are the central links in these gene networks. In vitro experiments showed that matrix metalloproteindase-12 (MMP-12) downregulation led to the decreased proliferation and invasion of bladder cancer cells and the phosphorylation level of extracellular regulated protein kinases(ERK). Conclusion Multiple bioinformatics analysis can identify the key genes in bladder tumors. CXCR4 and MAPK10 are the key links in the bladder gene network. MMP-12 is highly expressed in bladder cancer cells. Downregulation of MMP-12 can inhibit the proliferation and invasion abilities of bladder cancer cells through ERK pathway.

Objective@#To investigate the diagnostic value of serum levels of aquaporin-4 (AQP4) and matrix metalloproteinase-9 (MMP-9) in predicting malignant cerebral infarction in patients with cerebral infarction.@*Methods@#The clinical data about 66 patients with hemisphere massive cerebral infarction were collected. According to clinical symptoms and brain computed tomography (CT) findings, the patients were divided into malignant cerebral infarction group (MCI) and non-MCI group. The serum levels of AQP4 and MMP-9 were detected 1, 4, 7 days. The diagnostic value of AQP4 and MMP-9 in predicting occurrence of MCI was analysed by receiver operating characteristic (ROC) curve.@*Results@#The serum levels of AQP4 and MMP-9 in MCI group were significantly higher than those in non-MCI group in the fourth day (P<0.05). In MCI group, AQP4 and MMP-9 in the fourth day were significant higher than other two time points (P<0.05). But in non-MCI group, AQP4 and MMP-9 in different time points have no differences (P>0.05). ROC curve analysis showed that the aera under curve for the serum levels of AQP4 was 0.654 (95% CI: 0.516-0.792). When the serum AQP4=23.49 μg/L, the sensitivity and specificity were 0.57, 0.74, respectively.@*Conclusions@#Serum AQP4 and MMP-9 levels were increased in patients with MCI. Combined detection of serum AQP4 and MMP-9 is helpful for early differential diagnosis of MCI.

ABSTRACT

Objective@#To understand HIV/AIDS knowledge, attitude, and awareness of HIV/AIDS policies and regulations among vocational medical students in Sichuan, and to provide scientific basis for AIDS prevention and treatment in higher vocational medical colleges.@*Methods@#A total of 1 458 medical students in a vocational college in Sichuan Province were selected through stratified cluster random sampling and investigated with questionnaire on HIV/AIDS related knowledge, policies and regulations.@*Results@#The average score of AIDS knowledge was(6.63±1.31), and the awareness rate was 82.9%; The average score of attitude towards AIDS was(2.17±0.45), with the highest score being fear and avoidance(2.56±0.61), and the lowest score responsibility perception(1.66±0.58); The total score of HIV/AIDS related policies and regulations was(19.17±2.91). AIDS knowledge was positively correlated with attitude and understanding of policies and regulations(r=-0.24, 0.28, P<0.01) , the higher the knowledge score, the better the attitude and the higher the understanding of policies and regulations.@*Conclusion@#HIV/AIDS knowledge of higher vocational medical students has been improved, the fear and avoidance of AIDS is widespread. There are insufficient awareness in AIDS related policies and regulations. Higher vocational medical colleges should be more targeted AIDS health education.

ABSTRACT

The aim of this paper was to observe the combination therapy with total triterpenoids of Chaenomeles speciosa and omeprazole on indomethacin-induced gastric ulcer in rats, and explore its possible mechanism. Rats were randomly divided into normal group, model group, omeprazole monotherapy(3.6 mg·kg~(-1)) group, total triterpenoids of C. speciosa monotherapy(100 mg·kg~(-1)) group, total triterpenoids of C. speciosa and omeprazole combination therapy(100 mg·kg~(-1)+3.6 mg·kg~(-1)) group. Except for the normal group, the other groups were given indomethacin(20 mg·kg~(-1)) by oral once a day for 7 consecutive days. Then the treated groups were given corresponding drugs by gavage, once a day for 14 consecutive days. The next day after the last administration, half of the rats in each group were measured the gastric mucosal blood flow, gastric juice volume and serum TNF-α, IL-1β, IL-6, IL-4 and IL-10. After the remaining rats in each group were underwent pyloric ligation 4 hours after the last administration, the gastric endocrine volume, pH value and total acidity of gastric secretion were measured, then histological analysis was performed, MPO activity, cAMP content and histomorphological analysis were conducted. Real-time PCR was applied to detect the mRNA expressions of gastric tissue TNF-α,IL-1β, IL-6, IL-4, IL-10, VEGFA, A_(2A)R; the protein expressions of VEGFA, A_(2A)R, PKA, p-PKA, CREB, p-CREB, EGF, EGFR, p-EGFR, MUC6, TFF2 in gastric tissue were detected by Western blot. The results indicated that total triterpenoids of C. speciosa and omeprazole combination therapy might significantly increase gastric mucosal blood flow, gastric mucus volume, reduce gastric endocrine volume, secretion acidity and mucosal damage, decrease the levels of TNF-α,IL-1β and IL-6, increase the levels of IL-4 and IL-10 in blood and gastric tissue, inhibit the activity of MPO, increase the content of cAMP in gastric tissue, up-regulate the mRNA expressions of VEGFA, A_(2A)R and protein expressions of VEGFA, A_(2A)R, PKA, p-PKA, CREB, p-CREB, EGF, EGFR, p-EGFR, MUC6, TFF2 in gastric tissue, elevate p-PKA/PKA, p-CREB/CREB and p-EFGR/EFGR. Moreover, the combination therapy with total triterpenoids of C. speciosa and omeprazole was more obvious than those of two monotherapies. These aforementioned findings suggested that the combination therapy with total triterpenoids of C. speciosa and omeprazole on indomethacin-induced gastric ulcer have significant therapeutic effect on indomethacin induced gastric ulcer in rats, its mechanism might be related to regulating A_(2A)R/AKT/CREB, A_(2A)R/VEGFA, EGF/EGFR and MUC6/TFF2 signaling pathways, inhibiting pro-inflammatory factors, increasing gastric mucosal blood flow, up-regulating mucosal cell proliferation factors and promoting mucosal protective factors.
